In Wayne’s war pictures, the message was unmistakable: one has a duty to his country and he has to do his best to perform it. In the 1949 film Sands of Iwo Jima which earned him his first nomination as Best Actor, Wayne portrayed Sergeant Stryker, a professional soldier who fought in the blood-soaked beaches of Iwo Jima. He spent endless nights, and came up with the conclusion that the moon was actually made up of craters, mountains, and jagged surfaces. He announced to the public his discovery, and many people, including scientists, were outragged. Galileo was put on trial for wrongful thinking. People did not accept the fact that people were getting smarter, and the world was changing before their very eyes. Mankind is unreasonable, right before them ideas were being brought forward, and all they did was push them back down into the dirt. Pretty evil, the world was going to change anyway, it was not going to stay the same. It goes to show what mankind is also like. For example, if all of society is supposed to be equal, then why are nobles and the clergy only allowed to have important jobs, and the peasants forced to dig in the dirt for back breaking hours. He did not want to cause anymore trouble and see his father suffer, but he did agree to come home if he needs help, realizing that families are forever. Both Teddy and Glory will be there. â€Å"But the soul finds its own home if it even has a home at all† Glory, like Jack, is lost, alone and bitter. She carries on life with a heavy heart, as she takes care of her ailing father and a problematic brother. Feeling hopeless, she seeks attention and fortunately, her two brothers show it to her. She understands Jack, but not completely because of his secretive character and he, too, understood her, her feelings and battles. As the story progresses, there was a growing bond between brother and sister, whether it was taking care of their father or sharing thoughts and opinions, though avoiding to bring up the pressing issues amongst them. Nevertheless, they became better individuals because of each other. The Lord truly is wonderful! The story was ordered chronologically, with the occasional recalling of the past. The external problem was the coming home of Jack and Glory. He seemed distracted, causing worry to his sister and father and she, left unmarried, cheated, and alone. The narration of the story is character-based since the plot unfolds through the opinions and decisions made by the characters, not from having this setting to the next. No doubt that the story reflects religion, the uncertainty that everyone feels about it because there, as mentioned by Ames, is no absolute knowledge on what it really is. The stories among the characters parallel some characters and stories in the Bible such as David, Lazarus and The Prodigal Son. The plot evolved from the slight change in emotions. Glory began to feel needed and responsible in making her brother feel better and her father well, while Jack felt being part of the family. He saw and realized that his family would accept him for who he is and would try to do everything to make him feel welcome. In the outside, the plot was about the coming home and duties of both Glory and Jack, visiting of Ames and Teddy, and the parting of Jack. There were a lot of themes in the story such as guilt, loneliness, resentment and religion, but the main theme was the importance of the relationship among family members. It can help make everything much better, though slowly but surely. The theme is based from the belief of Robinson and I, definitely, agree with her. Family is the basic unit of society and is the foundation of every individual. The relationship among members should be strong in order for all of them to be stable and complete as each individual. Personal Application/Impression Home was a hard read. Though an avid reader, the book was more complicated and confusing compared to the fiction novels I normally read. The words have so much meaning and the plots were not written in a sequence, leaving me puzzled. I soon began to understand the context of the story and the meaning of those sentences that seemed too long after reading more. Marilynne Robinson writes Home in a reminiscing manner and with thorough explanations of the actions done by the characters. I have never read a novel as defined as this, enabling me to imagine the exact happenings in the story. Home has touched me, making me appreciate my family even more. I can feel the characters’ experiences when it comes to family, their effort to become closer and trying to offer help. It takes me from keeping my own problems to sharing it with my family because I now truly realize that they would accept me for who I am and help me with my troubles. The characters acted the way people actually do in real life and the settings were a possibility. Home truly is a beautiful and moving book, showing the importance of family. However; I did not enjoy the writing as much, though the storyline and intention of the author was wonderful. It was written with much effort, but a little too dragging and confusing. Maybe it was confusing because partly, I did not get to read the first book, Gilead, but the plots did jump from one to the other. Note that it is a must to read Gilead first for better understanding and appreciation of Home. The developing of the story was slow and some do enjoy that type of writing but I don’t so, it was a little dragging. It was hard to turn to the next page if the ending of a page is not as interesting. Robinson built a sturdy plot but bores some readers like me. It did not have action in every page but again, it is interesting for those who love to imagine being the character himself. A well-written novel, but no work of fiction is perfect for everybody.

Accurate results of total and ionic calcium level plays a pivotal role in patient care and management. Measurement of serum calcium helps to identify many clinical disorders. Many preanalytical conditions affect the blood calcium estimation. The aim of this study is to evaluate the effect of heparin, used as an anticoagulant on total blood calcium assay. Blood specimen from 10 healthy subjects were collected and the difference in total calcium contraction in plasma and serum samples was assessed. No significant changes were observed when plasma levels were compared with the serum values with p value of 0.56. Deming regression analysis yielded the equation: 1.02 Ãâ€" (serum total calcium value) + 0.28 mg/dl. The bias value was -0.109 mg/dl (95% CI: -0.248 0.030 mg/dl) for total calcium levels assayed using either tube was acceptable. Total calcium values did not significantly differ when either plasma or serum was used with the spearman correlation coefficients (r value) of 0.98. In conclusion, heparinized tubes can be used for the determination of total blood calcium. The use of plasma sample has an advantage in an emergency laboratory setting due to less processing time. Anticoagulants are additives that inhibit the clotting of blood; they are used only on ensuring that the concentration of the substance to be measured is changed as little as possible before the analytical process (3). The preanalytical laboratory testing takes two major steps: Blood collection and processing (4). The ionic calcium level in blood is recognized to be falsely decreased either due to dilution or binding effects of heparin, which is applied as an anticoagulant (5). The CSLI recommends the use of lyophilized calcium-balanced heparin syringes (6, 7) for collecting specimens for ionized calcium measurement. It has been proposed that ionized calcium levels can be affected by excess of heparin present in blood (8), although it was believed that the clinical significance was minimal (9). In this study, we are determining the effect of heparin on assay of total blood calcium to analyze preanalytical error in the collection of blood samples. Materials and Methods This study was approved by Swami Vivakanand Medical Mission ethical Committee. Informed consent was obtained from the donors. For evaluation of the effect of heparin on total blood calcium assay, blood samples were obtained from the 10 subjects. Blood samples were sequentially obtained by single venipunture from an antecubital vein in BD PresetR lithium heparin tubes (Ref. No. 367884) and BD vacutainer serum (Ref. No. 367812). All the subjects were between 18 50 years age group including both genders. Plasma levels of total calcium were compared to the serum calcium, which was used as a reference here. After the blood withdrawal, samples were then recapped into individual containers, followed by sample centrifugation. The plasma and serum samples were separated within 30 minutes of blood collection. Commercially available biochemical kit (Liquid Gold, Span diagnostic, India) by Arenazo-III method was used for total calcium Assay. Within 2 hours after collection, total calcium was me asured in all samples in triplicate and mean was used for further analysis. Data were evaluated using Analyse-it software (version 2.26). For all statistical comparisons, p values less than 0.05 were considered significant. Data concordance was evaluated via Deming regression analysis. The mean of differences (bias) and limits of agreement was calculated using the Bland and Altman method (10). Differences between total calcium level obtained from plasma and serum were compared using the paired t-test. Correlations were determined via calculation of Spearman correlation coefficients (r values).  ­Ã‚ ­Ã‚ ­Ã‚ ­ Results The total calcium values of samples analyzed in the present study ranged from 8.69 10.47 mg/dl and the mean total calcium values obtained using BD lithium Heparin was 9.34  ± 0.18 mg/dl whereas that for serum samples was 9.48  ± 0.18 mg/dl with SD of ≠¤0.57 (Table 1) by 1-way ANOVA. The Spearman correlation coefficient was r = 0.98 (p Discussion Serum is the preferred sample which is used by many laboratories for biochemical testing since it avoids the addition of anticoagulants that may interfere with some analytical methods or alter the true concentration of the parameters being measured. The use of serum or plasma in clinical pathology remains controversial. However, analyte stability is greater in serum than in plasma (11, 12) and differences between plasma and serum test results have often been documented (13). Moreover, in stored samples, the formation of fibrin strands is lower in serum than in plasma and therefore, there is lesser risk of occlusion in automated biochemical analyzers. However the use of plasma is preferred in some centres as it separates quickly saving preanalytical time thus reducing the total testing period (TTP); secondly, 15–20% more plasma sample can be obtained from the same volume of blood than the serum sample and lastly, with plasma there are no coagulation-induced changes or interferences (3). Heparin has been generally recommended as the most suitable anticoagulant for plasma collection (14) due to its physiological acceptance in vivo. Although in previous reports, a significant differences in selected parameters have been found between heparinized plasma and serum samples (13, 15). In our study, serum and heparinized plasma yielded similar results with no significant changes in total blood calcium assay this confirms the views of Thorensen, S.I.,et al (15) who also reported similar observations; and this adds up to the observation of Ladenson, J., et al (13) in which clinically significant changes were observed in ionic calcium observations on use of heparinized plasma over serum results.. We found that the Spearman correlation coefficient was 0.98, indicating a good correlation, when total calcium test results when plasma and serum were compared. Deming regression analysis yielded the slope of the regression line that did not approximate unity, suggesting that, in addition to the presence of a small constant bias, a small and non-significant proportional difference also existed between the test results obtained upon use of the two tube types (16). The total calcium assay bias was -0.109 mg/dl. The criteria of acceptable limits imprecision for total calcium ass ay is 1mg/dl and medically allowable error is 0.37 mg/dl (17, 18). In the present study, we observed that the total calcium concentrations of plasma samples were slightly lower than serum samples with no clinically significant differnece and the bias values were smaller than US CLIA 1988 targets. Although collection of blood samples from the patients administered with heparin prior to the collection may hold in excess of heparin, increasing clotting time in the collection tube and thereby increasing the potential for the establishment of latent fibrin in the preanalytical phase. Preanalytical variables associated with blood collection should be further standardized to ensure the accuracy of test results. It is impractical to expect tube manufacturers to test out their subway systems on all possible assay platforms; this is a project for individual research laboratories. Conclusion We conclude that heparinized tubes are suitable for sample collection for total calcium assay. With the use of heparin tubes use faster processing of blood samples can be possible, which is valuable in an emergency setting and in intensive care units. Moreover, single sample can be used for multiple purpose, including both hematological and biochemical analyses. Brain Drain One of the major concerns of today's companies is the shortage of labor, especially in management. The baby boomers are nearing retirement age in the United States and the birth rate is dropping. These circumstances, coupled with the booming economy are the main causes of the labor shortage. There is a high demand for labor but the once seemingly bottomless pool of employees and managers that companies drew from has started to dry up. What are the factors that contributed to the problem and how are today's corporations going to handle this problem? The type of labor needed in today's society has been undergoing a constant change. There is an increase in demand for workers but there is a much greater demand for educated white-collar workers, especially management material. Projections state that the growth in managerial positions will increase 20% by the year 2010 yet the population aged 35-50 will decrease nearly 10%. What these figures say is the already diminishing supply of executives is going to dwindle even more over the next 10 years. There is a shortage of blue-collar workers now and there will also be an even greater shortage of them in the future. In order for employers to find people who are willing to perform unskilled, repetitive jobs they are going to have to be willing to raise the level of compensation offered to employees. If McDonalds needs someone to flip hamburgers they better be prepared to pay double to triple minimum wage. There are a wide variety of employment opportunities and today's workforce can afford to be selective when choosing a job. The demand for employees is high while the supply is low. The figures on the change in average population ages and growth in industrialized nations is beginning to make the corporate world stand up and take notice. If the trends continue as they have been for the past thirty years, the shortage of labor is going to continually get worse with each year that passes. The predictions from the United States Census Bureau state that between 1990 and 2000 the increase of the American population over 60 will be 10.5% but in 2010 to 2020, the increase will be 32.5%. The change in the 60 plus population in the United States is projected to nearly triple in thirty years. Compare these figures to the increase in under sixty-year-old population. I think that they are moving in the right direction. Offering flexibility in what was once a very rigid structured hierarchy is certainly a step in the right direction. A rewarding profession is not exclusively defined by the income generated anymore, rewarding can also mean personally rewarding. A good employee doesn't necessarily work 40 hours each and every week, some work part time or are involved in a job-sharing program. In order to keep the workforce meeting the needs of the economy, the employer must continue to become more flexible and remain open to suggestions. Today's companies are on the right track. I believe that with proper management and forward thinking, we will be able to cope with the ever-increasing shortage of labor. Our ancestors first cultivated plants some ten thousand years ago. They domesticated animals later and then selectively bred both plants and animals to meet various requirements for human food. Humans discovered natural biological processes such as fermentation of fruits and grains to make wine and beer, and yeast for baking bread. Manipulation of foods is not a new story, therefore. The latest agricultural discovery uses genetic engineering technology to modify foods. Farmers and plant breeders have been changing crop plants to improve characteristics such as size, resistance to disease and taste. Plants which grow well, have a higher yield or taste better are selected and bred from. This is still the most widely used technique for developing new varieties of a crop, and is limited by natural barriers which stop different species of organisms from breeding with each other. Genetic modification is very different to these traditional plant breeding techniques. Genetic modification is the insertion of DNA from one organism to another, usually by molecular technologies. Genetically Modified Foods (GMF) are animals or plants that have had genetic modification. This changes the characteristics of the organism, or the way it grows and develops. The famous frost-resistant tomato example is perfect in illustrating this point. With a tomato that  resists frost, the season for growing them would be longer and therefore a farmer would be able to produce more tomatoes in one year than they were able to do in the past. Gene technology not only gives us the potential to select the exact characteristics we want in an organism, but it also enables us to cross species barriers. For example, we can take an insecticide-producing gene from a bacterium and insert it into a plant, making the plant resistant to insect attack. This new-found ability to cross species barriers is what makes gene technology such a powerful tool. Producing enough food for the world’s population without using up all the available land is an enormous challenge. One solution is to develop crops that yield more with fewer inputs; that are more resistant to diseases; that spoil less during storage and transport; that contain more useful nutrients; and that can grow in agricultural land that has been degraded. Gene technology gives us the potential to do this. Genetically modified foods have been available since the 1990s. The principal ingredients of GM foods currently available are derived from genetically modified soybean, maize and canola. The first commercially grown genetically modified food crop was a tomato created by Calgene called the FlavrSavr. Calgene submitted it to the U.S. Food and Drug Administration (FDA) for assessment in 1992; following the FDA’s determination that the FlavrSavr was, in fact, a tomato, did not constitute a health hazard, and did not need to be labeled to indicate it was genetically modified, Calgene released it into the market in 1994, where it met with little public comment. Considered to have a poor flavor, it never sold well and was off the market by 1997. However, it had improved solids contents which made it an attractive new variety for canned tomatoes. Transgenic crops are grown commercially or in field trials in over 40 countries and on 6 continents. In 2000, about 109.2 million acres (442,000 km ²) were planted with transgenic crops, the principal ones being herbicide- and insecticide-resistant soybeans, corn, cotton, and canola. Other crops grown commercially or field-tested are a sweet potato resistant to a US strain of a virus that affects one out of the more than 89 different varieties of sweet potato grown in Africa, rice with increased iron and  vitamins such as golden rice, and a variety of plants able to survive extreme weather. Between 1996 and 2001, the total surface area of land cultivated with GMOs had increased by a factor of 30, from 17,000 km ² (4.2 million acres) to 520,000 km ² (128 million acres). The value for 2002 was 145 million acres (587,000 km ²) and for 2003 was 167 million acres (676,000 km ²). Soybean crop represented 63% of total surface in 2001, maize 19%, cotton 13% and canola 5%. In 2004, the value was about 200 million acres (809,000 km ²) of which 2/3 were in the United States. In particular, Bt corn is widely grown, as are soybeans genetically designed to tolerate glyphosate herbicides. Future applications of GMOs include bananas that produce human vaccines against infectious diseases such as Hepatitis B, fish that mature more quickly, fruit and nut trees that yield years earlier, and plants that produce new plastics with unique properties. The next decade will see exponential progress in GM product development as researchers gain increasing and unprecedented access to genomic resources that are applicable to organisms beyond the scope of individual projects. Biologist Stephen Nottingham explains the risks of GMF:“Experimental trials with transgenic organisms are usually conducted strict regulations to minimize the potential spread of genetic materialÂ…Even given these regulations, however, no field trial can be said to be 100% secure. For instance, if an individual consumer who is allergic to broccoli eats a banana that just happens to have a little broccoli DNA under the peel, that person might get sick. Some studies on animals indicate that consuming genetically-modified foods may cause allergic responses, compromise immune systems and inhibit organ growth, although no proven cases of widespread reactions have been definitively documented. Opponents of biotech foods want other questions answered, as well. Will re-engineering a plant or animal to serve a specific end, such as improving taste, decrease its nutritional value? Will consuming genetically-modified food products make a person more resistant to antibiotics, which are widely used to treat bacterial infections? Does consuming milk or meat from livestock that has been injected with growth hormones (a form of biotechnology that is different from genetic modification) subject consumers to early puberty, cancer, and other ailments?Since neither side has been able to provide definitive answers, the jury is still out on food safety; after all, genetic technology itself is barely decades old. So one can condense the issue into a single question: should we move forward with new technologies that might help provide higher crop yields, new and interesting types of food products, and more profits for the companies that own the technology; or play it safe and wait until we better understand the health and environmental consequences of manipulating life forms that took generations to develop?Multinational Corporations benefit because GMF can be very profitable. GMF have taken hold quickly because multinational corporations with the resources to make large financial investments in research and development can profit directly. Multinational companies can spread out the benefit and profit to many branches of their businesses. Many such corporations combine the following: an agrochemical company, a seed  company, a pharmaceutical company, a food processing company and sometimes businesses involved with veterinary products. Developments in one part of the corporation can be used t o sell products in another branch. Farmers benefit in the short term because they can grow and sell more crops with fewer problems due to weeds, pests, fungi or frost. The genetically modified seed is designed to resist these traditional enemies. Food processing companies benefit from a ready supply of raw food ingredients designed for specific processing needs. Genetically modified tomatoes and potatoes, for instance, have higher solid contents and yield more sauces and French fries. These foods take longer to ripen and rot. Thus less food is spoiled and more gets processed. Supermarkets benefit for the same reasons. The fresh produce lasts longer on the shelves and is more profitable. Consumers, to date, haven’t benefited. GMF have been developed for the convenience of the producer and processor. Yet they cost more to produce and the costs get passed along to the consumer. Eventually there will be some kind of designer novelty foods for shoppers to try. Nottingham adds that there are many other concerns including ethical questions involving animal welfare, whether DNA is actual life, and intellectual property rights and genetic resources from the Third World. (Bragi)The world’s poorest nations account for around 95.7% of the world’s genetic resources. Traditional farming practices involve farmers retaining seeds, from the harvest of one year’s crop, for planting in the following year. This practice saves money on buying seed and in itself represents a continuous selection for yield and resistance to pests and diseases. However, with genetically modified seed, royalties are payable to the companies holding the patent for the seed. Under world trade agreement rulings, farmers have to make substantial royalty payments to multinational companies if they keep seed for replanting, even if the crop happens to be native to their particular country. Genetic engineering is a valuable new technology that can develop more plentiful and nutritious foods, with great potential benefits for humanity and the environment, and this new scientific discovery needs to be implemented as quickly as possible for humanitarian reasons. As with every new scientific technology, harmful side effects of genetic engineering are inevitable and great care should be taken in its implementation, including carefully controlled long-term tests on human health and environmental impacts. All genetically engineered foods have been thoroughly tested and demonstrated to be safe before they are released into the marketplace. However, this testing is typically conducted only on rats and other animals, by the companies involved. Very little of this research has been reviewed by independent scientists and then published in scientific journals. Genetically engineered foods are usually â€Å"substantially equivalent† to other foods, with no increased risk to human health, and no need for the lengthy and expensive human testing demanded of, for example, new food additives. However, the unpredictable disruptions in normal DNA functioning caused by genetic engineering can produce unanticipated and unknown side effects for human health, including unknown and unpredictable toxins and allergens, and these possibilities can only be definitively assessed through human testing. Genetic engineering is a scientific and technological process, and its evaluation and governmental regulation should be based on purely scientific and objective criteria. To have a purely scientific evaluation of genetically engineered foods, we need more science, especially human studies and environmental studies. Moreover, purely scientific assessment of genetic engineering ignores the fact that, for many people, food has cultural, ethical and religious dimensions that must also be considered. Alan McHughen, author of Pandora’s Picnic Basket: The Potential and Hazards of Genetically Modified Foods, in the introduction he states:†Make no mistake: I am in favor of an orderly and appropriately regulated introduction of some GMOs into the environment and marketplace, and I  adamantly oppose others. There are good reasons to ban certain products of genetic technology, and good reasons to allow, with management, certain others; some may require no extraordinary regulation at all. If your opinion differs from mine after reading this book, I hope you will be able to justify, if only to yourself, why we disagree. My philosophy is to be skeptical, be critical, even cynical of claims by business interests, government agencies, and activist groups. But also keep an open mind and then decide for yourself.† (Internet 7)ThereÂ’s no doubt that the GM food supply should be closely monitored and regulated, but that doesnÂ’t mean it should all be banned. I believe that genetic engineering of plants, animals, and humans has much to offer as long as we are aware of potential benefits and side effects.
